Resumo:Nature represents the main source of organic compounds, many of which have significant added value due to their remarkable potential for therapeutic and industrial applications. Chitosan, for example, is a natural polysaccharide derived from chitin, abundant in crustacean exoskeletons. Its importance is due to its biocompatible, biodegradable and non-toxic properties, which makes it valuable in medical formulations, such as dressings, controlled drug release devices and regenerative therapies. In turn, naphthoquinones, a class of aromatic chemical compounds, have a wide range of biological activities that include antiviral, anticancer, antimicrobial and anti-inflammatory properties. These characteristics give them significant relevance in the search for new drugs and therapeutic agents. In this study, the naphthoquinones 3-chloromethylene-menadione (NQ1) and 2,3-dichloro-1,4-naphthoquinone (NQ2) were encapsulated in chitosan nanocapsules (CNP) by the ionotropic gelatinization technique. Homogeneous preparations of chitosan nanocapsules of 193.5 ± 1.9 and 193 ± 9.3 nm sizes, Z potential of +30.6 to +33.1 mV, loaded with NQ1 (CNP-NQ1) or NQ2 (CNPQ-NQ2), respectively, were characterized by DLS, FTIR, SEM, TGA, DSC methods. With encapsulation efficiencies ≥ 96%, the water solubility of both naphthoquinones could be improved, making them suitable for application in biological studies. Both CNP-NQ1 and CNP-NQ2 retained the naphthoquinones antimicrobial activity against Staphylococcus aureus and Staphylococcus epidermidis, and, although they exhibited low toxicity against healthy human cells (HFF-1), only CNP-NQ1 showed adequate therapeutic index > 10, ensuring its safe application to human tissues. Furthermore, CNP-NQ1 did not impair the migration of healthy human fibroblast cells in scratch assays, adding to the novel formulation promissory wound healing properties. The findings reported here emphasize that CNP-NQ1 may be useful in protecting injured tissue from bacterial contamination, commonly associated with skin infections, not only by reducing the bacterial cargo but even in the healing process until the complete recovery of the tissue.
